Requirement of Fc-Fc Gamma Receptor Interaction for Antibody-Based Protection against Emerging Virus Infections

چکیده

Identification of therapeutics against emerging and re-emerging viruses remains a continued priority that is only reinforced by the recent SARS-CoV-2 pandemic. Advances in monoclonal antibody (mAb) isolation, characterization, production make it viable option for rapid treatment development. While mAbs are traditionally screened selected based on potency neutralization vitro, clear additional factors contribute to vivo efficacy mAb beyond viral neutralization. These include interactions with Fc receptors (FcRs) complement can enhance neutralization, clearance infected cells, opsonization virions, modulation innate adaptive immune response. In this review, we discuss studies, primarily using mouse models, identified role Fc-Fc?R optimal antibody-based protection virus infections.

Fc gamma receptor glycosylation modulates the binding of IgG glycoforms: a requirement for stable antibody interactions.

FcγRs play a critical role in the immune response following recognition of invading particles and tumor associated antigens by circulating antibodies. In the present study we investigated the role of FcγR glycosylation in the IgG interaction and observed a stabilizing role for receptor N-glycans. We performed a complete glycan analysis of the recombinant FcγRs (FcγRIa, FcγRIIa, FcγRIIb, FcγRIII...

A novel role for the Fc receptor gamma subunit: enhancement of Fc gamma R ligand affinity

The Fc receptors (FcR), which belong to the immunoglobulin (Ig) superfamily, bind to specific Ig isotypes with varying affinities triggering complex immune defense responses. Several of the FcR that lack signaling motifs in their cytoplasmic domains rely on associated subunits to transmit signals. Two classes of FcR that bind the Fc portion of IgG, Fc gamma RI, and Fc gamma RIIIa associate with...

Ligation of Fc gamma receptor IIB inhibits antibody-dependent enhancement of dengue virus infection.

The interaction of antibodies, dengue virus (DENV), and monocytes can result in either immunity or enhanced virus infection. These opposing outcomes of dengue antibodies have hampered dengue vaccine development. Recent studies have shown that antibodies neutralize DENV by either preventing virus attachment to cellular receptors or inhibiting viral fusion intracellularly. However, whether the an...

Neuraminidase augments Fc gamma receptor II-mediated antibody-dependent enhancement of dengue virus infection.

Antibody-dependent enhancement (ADE) of dengue virus infection occurs when neutralizing antibodies at sub-neutralizing concentrations or non-neutralizing antibodies form complexes with the virus. These virus-antibody complexes can then attach to a Fc gamma receptor-bearing cell, via the Fc portion of the immunoglobulin, resulting in an increased number of infected cells. ADE may be responsible ...

Antibody- and Fc-receptor-based therapeutics for malaria.

Abs (antibodies) are complex glycoproteins that play a crucial role in protective immunity to malaria, but their effectiveness in mediating resistance can be enhanced by genetically engineered modifications that improve on nature. These Abs also aid investigation of immune mechanisms operating to control the disease and are valuable tools in developing neutralization assays for vaccine design. ...
